Most of the VECTASTAIN® ABC kits, ImmPRESS™ Reagents, and Vector’s other enzyme and fluorescent-based detection products can be easily adapted for use on automated staining instruments that allow user-supplied reagents. Many of the detection systems offered in this catalog can produce the same high sensitivity and background-free, specific staining results with these automated instruments as those obtained using manual staining techniques.
In addition to greatly expanding the number of detection reagent choices and antigen visualization preferences, the use of Vector® reagents may result in substantial cost savings. These savings can result from the purchase of concentrated reagent kits or bulk reagents. Indirect savings may also be realized by an ability to further dilute expensive primary antibodies.
The VECTASTAIN®ABC Kits, the VECTASTAIN® ABC-AP Kits, and the respective Vector® enzyme substrate kits are popular choices for use on automated staining machines. Both the concentrated forms and Ready-To-Use products can be used for standard, programmed, automated staining in same day procedures. For overnight applications, VECTASTAIN® Ready-To-Use (R.T.U.) products are recommended to achieve optimal staining. Substrates should be prepared fresh before use.
The VECTASTAIN® kit or other reagent selection is generally the same for manual or automated systems. In most cases a slight modification of the diluted working solution, such as an increase or decrease in concentration or the addition of a detergent, is all that is required to adapt Vector® detection reagents to achieve sensitive, background-free staining. Most products as supplied are used in a straightforward manner with no adjustments to the recommended procedural order, incubation times or temperatures.
The ImmPACT™ peroxidase substrates are an ideal choice for automated staining. High sensitivity and stability of the working solutions allow faster staining on lower primary antibody concentrations and optimal substrate performance.
Several of our accessory items can also be used with an automated staining protocol. These include the Antigen Unmasking Solution, normal sera and bovine serum albumin (BSA) for blocking non-specific binding, blocking kits for eliminating biotin/(strept)avidin interactions with tissue elements, and counterstains for highlighting morphological features. Mounting media for both fluorescent or enzyme detection systems are also available.
